盘州大厂茶(Camellia tachangensis)的生物碱类、儿茶素类及氨基酸类组成特性分析
Composition characteristics of alkaloids,catechins and amino acids in Panzhou Camellia tachangensis

Abstract
[Objective]The biochemical composition characteristics of Panzhou Camellia tachangensis was explored to provide the scientific basis for system identification of wild tea trees and new variety breeding,genetic resource conservation and utilization of characteristic Panzhou C.tachangensis in Guizhou.[Method]29 biochemical components of 25 Panzhou C.tachangensis germplasms and 3 control varieties were de-tected by high performance liquid chromatography(HPLC).The composition characteristics and biochemical quality of Panzhou C.tachangensis were comprehensively evaluated by a set of multiple statistical methods including orthogonal partial least squares discrimination analysis(OPLS-DA),correlation analysis,principal component analysis and cluster analysis.[Result]There was an abundant genetic diversi-ty in Panzhou C.tachangensis germplasms.The variable coefficient of 28 biochemical components was 0.79%(GCG)-241.4%(GCU)with an average of 46.08%.The genetic diversity index was 0.861(GCU)-2.001(EC)with an average of 1.798.Theacrine,ECG and theanine were the dominant alkaloids,catechins and amino acids in Panzhou C.tachangensis germplasms respectively.The OPLS-DA analysis showed that 11 biochemical components with VIP>1 of caffeine,EGCG,theacrine,cysteine,proline,GCG,methionine,ECG,EGC,theanine and EC could be used to identify Panzhou C.tachangensis and cultivated tea varieties.There was the certain regularity in the correlations be-tween 28 biochemical components.The correlation between alkaloids and amino acids was higher than between alkaloids and catechins.There was a certain cooperativity and antagobism between catechins.EC was negatively related to EGCG and GCG significantly but EDCG was posi-tively related to EGC and ECG very significantly.There were higher positive correlations between different amino acids mostly.There were the positive correlations between proline,tyrosine,lysine,isoleucine,leucine and phenylalanine very significantly.The accumulation contribution rate of 8 principal components selected by principal component analysis reached 82.834% and the first principal components of isoleucine,leucine,serine,lysine,phenylalanine and proline were the key factors to evaluate the biochemical quality of Panzhou C.tachangensis.25 Pan-zhou C.tachangensis germplasms could be divided into 2 groups by cluster analysis.Group A included 8 Panzhou C.tachangensis germplasms of PZ-01,PZ-02,PZ-03 and so on.Group B included other 17 Panzhou C.tachangensis germplasms.The content of theacrine and arginine in Group A was significantly higher than that in Group B and the content of serine,threonine,proline,tyrosine,lysine,isoleucine,leucine and phenylalanine in Group A was higher than that in Group B very significantly.[Conclusion]The theacrine,EC,ECG and methionine in Pan-zhou C.tachangensis germplasms with a biochemical specificity are peculiar high accumulative biochemical components.Amino acids are the keys to evaluate the biochemical quality of Panzhou C.tachangensis germplasms and to cause internal differentiation of Panzhou C.tachangen-sis germplasms.PZ-01,PZ-02,PZ-06,PA-09 and PZ-16 in Panzhou C.tachangensis germplasms have the application value in breeding of specific tea varieties.关键词
