天津地区芹菜软腐病病原菌的生物学特性和抗病品种初步筛选
Biological Characteristics of the Pathogen Causing Celery Soft Rot and Preliminary Screening of Disease-Resistant Varieties

Abstract
To elucidate the growth characteristics of the causal pathogen and conduct preliminary screening of resistant cultivars in Tianjin,samples were collected from celery fields with recurrent soft rot outbreaks in Xinkou Town,Xiqing District.Pathogenic bacte-ria were isolated using a gradient dilution method and identified as Pseudomonas sp.through 16S rDNA sequencing analysis.Growth characterization revealed that the optimal carbon and nitrogen sources for the representative strains TX09 and TX61 were yeast extract and peptone,respectively,with an optimal growth temperature of 37 °C.Pathogenicity assays demonstrated that both strains induced symptoms consistent with celery soft rot in oilseed rape(Brassica napus)and cucumber(Cucumis sativus),but failed to infect tomato(Solanum lycopersicum),melon(Cucumis melo),or carrot(Daucus carota).Upon infection,peroxidase(POD)and catalase(CAT)activ-ities in celery tissues significantly increased,while superoxide dismutase(SOD)activity transiently decreased followed by an increase.Notably,preliminary screening of resistant cultivars identified complete immunity in the French celery cultivar and significant resis-tance in the Ventura(WTL-light)cultivar.In summary,this study confirmed that the pathogen of celery soft rot in Tianjin was a bac-terium belonging to the genus Pseudomonas.The biological characteristics of the pathogen were systematically characterized,and dis-ease-resistant celery varieties were preliminarily screened.These findings provide a scientific basis for the diagnosis and management of this disease.关键词
