Abstract

In order to explore the molecular mechanism of proliferation and differentiation of bovine skeletal muscle satellite cells and search for skeletal muscle associated genes in different periods,this study conducted transcriptomic sequencing analysis of bovine skeletal muscle satellite cells at proliferation and differentiation stages,and used bioinformation technology to analyze and screen functional genes.The results showed that a total of 645 differentially expressed genes were obtained,including 388 up-regulated genes,most of which were related to muscle fiber development.There were 257 down-regulated genes,mostly genes related to DNA synthesis.The difference level of differentially expressed genes,GO analysis,KEGG analysis and PPI protein interaction analysis showed that MYL1,MYH3,TNNC2,TNNT3 and TNNI2 were important candidate genes affecting muscle development in bovine skeletal muscle satellite cells.CDC6,KIF2C,PLK1,KIF11,MCM3,MCM4,MCM5 and MCM7 were important candidate genes af-fecting DNA replication in bovine skeletal muscle satellite cells.In this study,5 differentially expressed genes were screened out,which were involved in biological processes such as skeletal muscle contraction.8 differentially expressed genes were screened during proliferation,which were involved in biological processes such as DNA unwinding and DNA replication initiation.The above genes may play an important role in the proliferation and differentiation of skeletal muscle satellite cells and lay a theoretical foundation for beef cattle breeding.
