KIF14对黏着斑蛋白SVIL在细胞分裂沟中定位的作用
Effect of KIF14 on the localization of adhesion plaque protein SVIL in the cleavage furrow of cell
摘要
Abstract
To elucidate the relationship between kinesin KIF14 and adhesion plaque protein supervillin(SVIL)during cytoki-nesis,HeLa cells transfected with siRNA targeting KIF14(si-KIF14)and SVIL(si-SVIL)were utilized as experimental models.The expression levels of KIF14 and SVIL were assessed using Western blot.Immunofluorescence staining was employed to specifically label KIF14 and SVIL,and their precise localization during cytokinesis was observed through super-resolution con-focal fluorescence microscopy.The results demonstrated that the knockdown of KIF14 did not affect SVIL expression;however,SVIL was unable to localize to the cleavage furrow.In contrast,the knockdown of SVIL had no effect on KIF14 expression or its localization to the cleavage furrow.These findings indicate that SVIL localization to the cleavage furrow is dependent on KIF14,highlighting the crucial role of KIF14 in the process of cytokinesis.关键词
