NaCl Stress Resistance Evaluation of 232 Rapeseed Germplasms during Germination and Identification of Responsive Genes

Abstract

To screen NaCl-tolerant rapeseed germplasms and analyze the potential mechanisms of NaCl stress response at the transcriptional level,the NaCl resistance of 232 rapeseed germplasms dur-ing seed germination was evaluated.Ten NaCl-tolerant germplasms,including NWAFU114,and twelve NaCl-sensitive germplasms,including NWAFU99,were identified.Furthermore,we demonstra-ted that NWAFU114 enhanced NaCl tolerance during germination by promoting antioxidant enzyme activity and alleviating membrane damage.Using the identified NaCl-tolerant germplasms and NaCl-sensitive germplasms,a transcriptome analysis was performed with NaCl-treated germinating seeds cultured six hours under light condition after three days of stratification.A total of 12 135 differential-ly expressed genes(DEGs)were identified.Gene ontology(GO)analysis showed that among signifi-cantly enriched DEGs,many were involved in biological processes including responses triggered by ab-scisic acid(ABA),jasmonic acid(JA),oxidative stress,osmotic stress,and cell wall biogenesis,all of which are known to play an important role in plant stress responses.
