Metabolite Differences between Rough-skinned and Normal Korla Pears

Abstract
This studyinvestigates metabolite difference between rough-skinned and normal Korla pears during the ripening stage of Korla fragrant pears to enhance its quality,and to provide a theoretical basis for improving fruit quality and promoting the development of Korla fragrant pear products.Differences in fruit quality were distinguished through visual and physiological assessments.The me-tabolites were analyzed using ultra-high performance liquid chromatography tandem mass spectrome-try(UPLC).The data were processed by Principal Component Analysis(PCA),Orthogonal Partial Least Squares Discriminant Analysis(OPLS-DA),and clustering analysis.The combination of fold change and the OPLS-DA model VIP score were used to identify differential metabolites.A total of 815 metabolites were detected in Korla fragrant pear,which were categorized into 11 groups,inclu-ding flavonoids,phenolic acids,lipids,and terpenes,among these,flavonoids,phenolic acids,and lipids accounted for 52%of the total metabolites.A total of 163 different metabolites were identified in fruits of different quality.In rough-skinned pears,53 metabolites,including amino acids(8/9),or-ganic acids(8/9),and alkaloids(5/6),were found to be abundant and had high concentrations.In normal pears,114 metabolites,including flavonoids(56/64),phenolic acids(26/32),lignans,couma-rins(6/7),tannins(5/5),and quinones(2/2),were abundant.KEGG pathway enrichment analysis revealed that the differential metabolites were distributed in 51 metabolic pathways,with significant enrichment in flavonoid and flavonol metabolism,as well as flavonoid metabolism(P<0.05).In nor-mal pears,eight types of flavonoids and flavonols were found in higher concentrations,while in rough-skinned pears,four types of alkaloids were present in higher concentrations,mainly synthe-sized through the metabolic pathways of arginine and proline.These findings suggest that the differ-ence in the quality of Korla fragrant pears may be related to the metabolism of flavonols and fla-vonoids,and synthesis pathways of arginine and proline.关键词
