Abstract

Bovine viral diarrhea virus(BVDV)may cause mucosal disease,respiratory and gastrointestinal tract infections,and reproductive failure in cattle,which have resulted in significant economic losses in global cattle industry.To establish and verify a fluorescence quantitative PCR method for rapid detection of BVDV,primers and Taqman probe were designed for BVDV 5'-UTR sequence,and the reaction system was optimized.The specificity,reproducibility and sensitivity of the fluorescence quantitative PCR method for BVDV detection was tested.This method showed no cross reactivity with bovine enterovirus(BEV),bovine rotavirus(BRV)and bovine parvovirus(BIPV),indicated good specificity.This method had a good linear relationship in the range of 1×102~1×107 copies/μL,R2=0.996 7.The lowest detection limit was 1×102 copies/μL.The intra-and inter assay coefficient of variation(CV)were<2%,suggested the excellent repeatability and stability.The developed Taqman fluorescence quantitative PCR method can be used for the rapid diagnosis of BVDV,thus facilitating the prevention and control of BVDV.
