A Novel Wall-breaking Technique of Preserving Eimeria tenella Intact Shape

Abstract
The aim of this experiment was to investigate the wall-breaking and dyeing techniques for Eimeria tenella(E.tenella)protoplast in their entirety.After pretreating the immature E.tenella oocysts with a freeze-thaw cycle and a concentrated hydrochloric acid-ethanol mixture,a dye known as Hoechst 33258 was added to fluorescently stain the E.tenella protoplast.The results revealed that the E.tenella oocysts pretreated with both the freeze-thaw cycle and the concentrated hydrochloric acid-ethanol mixture were significantly intact,and the cell nucleus of E.tenella was strongly stained,which was not observed in the control group or the group that only used the freeze-thaw cycle technique.This finding indicated that the tissue structure of the E.tenella oocysts was disrupted,allowing the dye Hoechst 33258 to penetrate through the oocyst wall and cell membrane,resulting in the fluorescent staining of the E.tenella protoplast.Through this experiment,a novel and original wall-breaking technique was developed to maintain the integrity of the immature E.tenella oocysts,for the first time,dyeing of E.tenella protoplast was achieved.Marking and tracing the coccidian oocysts allows a direct observation of their morphological changes and quantitative changes,which can help better understand the pathogenesis of coccidiosis and provide new ideas for developing anti-coccidial drugs.关键词
