Cloning of Protein Phosphatase EsPP2C48 Gene of Elymus sibiricus and Analysis of Its Response to Drought Stress

Abstract
The protein phosphatase 2C(PP2C)family plays an important role in plant responses to abi-otic stress.In this study,the EsPP2C48 gene was successfully cloned from Elymus sibiricus cv.Zhongcao No.28 by PCR,using InterPro,NetPhos 3.1 and STRING bioinformatics analysis tools,the structure and function of the protein encoded by the EsPP2C48 gene were analyzed.The results indicate that EsPP2C48 was a hydrophilic protein without signal peptides and transmembrane domains,which was mainly located in the cytoplasm and contains a PPM-type phosphatase domain.Phosphorylation site prediction showed that there were multiple potential regulatory sites.EsPP2C48 mainly interacts with proteins of the mitogen-activated protein kinase(MAPK)family in serine/threonine kinases.Taking 0 hours of drought stress as the control,the expression level of EsPP2C48 gene in leaves showed an up-regulated trend under drought stress,which was significantly higher than that of the control at 2 h(P<0.05),reached the peak at 6 h,and then began to decline,the expression of this gene was significant higher than that of the control at 12 h(P<0.05),but not significantly higher than that of the control at 24 h and 48 h.The expression level of EsPP2C48 in the roots of Elymus sibircus was down-regulated as a whole,and up-regulated slightly after 2 h and 4 h of stress,but there was no significant difference compare with the control,after 12 h stress,the expression level was significantly lower than that of the control(P<0.05).In conclusion,EsPP2C48 gene plays an important regulatory role in the response of Elymus sibiricus to drought stress.关键词
