摘要

Abstract

The gastrointestinal microbiota is known as the host's symbiont and an important functional organ,playing a key role in animals and providing a new perspective for studying the adaptive evolution of animals.To explore the en-ergy metabolism mechanism of Tibetan sheep adapting to high-altitude environments,this study selected 6 Tibetan sheep and 6 Hu sheep ewes of similar weight under the same feeding management conditions,and used 16S rRNA,gas chromatography,and quantitative PCR to measure the structure of the rumen microbial community,the concentration of volatile fatty acids(VFAs),and the expression level of VFAs absorption and transport-related genes in Tibetan sheep and Hu sheep.The results showed that the abundance of Bacteroidetes and Rikenellaceae_RC9_gut_group in the rumen of Tibetan sheep was significantly higher than that of Hu sheep(P<0.05),which increased the levels of carbohydrate metabolism,lipid metabolism,and protein turnover,enabling Tibetan sheep to effectively ferment and utilize forage and absorb and metabolize VFAs;the total concentration of VFAs in the rumen of Hu sheep was significantly higher than that of Tibetan sheep(P<0.05),and the contents of acetic acid,propionic acid,and pentanoic acid were all higher than those of Tibetan sheep,and the differences were significant(P<0.05),but the contents of isobutyric acid and isova-leric acid were significantly lower than those of Tibetan sheep(P<0.05).The expression levels of Down Regulated in Adenoma,Anion Exchanger 2,Na+/H+exchange 1 and Na+/H+exchange 2,which are related to VFAs transport in the ru-men epithelium of Tibetan sheep,were all significantly higher than those of Hu sheep(P<0.05),indicating that Tibetan sheep can more effectively use VFAs for energy,so having a unique rumen microbial community and stronger rumen fermentation function is a characteristic and strategy for Tibetan sheep to adapt to the high-altitude environment.
