不同浓度果糖与葡萄糖对湖羊精液低温保存效果的影响
Effects of Different Sugars on the Cryopreservation of Semen from Hu Sheep

Abstract
In order to study the effects of different concentrations of fructose and glucose on the cryopreservation of Hu sheep semen,different concentrations(0 g/mL,0.01 g/mL,0.02 g/mL,0.03 g/mL,0.04 g/mL)of fructose and glucose were added to the basic diluent of Hu sheep semen for cryopreservation.The optimal concentration of fructose and glu-cose for the cryopreservation dilutions of Hu sheep semen was determined by measuring sperm viability at different preservation time(0-5 days).Based on this concentration,the two sugars were added individually and jointly to create six different concentration treatment groups(0.02 g/mL fructose,0.02 g/mL glucose,0.01 g/mL fructose+0.01 g/mL glucose,0.02 g/mL fructose+0.02 g/mL glucose,0.005 g/mL fructose+0.015 g/mL glucose,and 0.015 g/mL fructose+0.005 g/mL glucose)for cryopreservation of semen from Hu sheep.The results indicated that at 1-5 days of preserva-tion,the 0.02 g/mL fructose treatment group exhibited the highest sperm viability,vigor,linear motion rate(VSL),cur-vilinear motion rate(VCL),and mean path rate(VAP)in Hu sheep.From 2-5 days of preservation,the plasma mem-brane integrity rate and acrosome integrity rate of Hu sheep spermatozoa in the 0.02 g/mL fructose-treated group were significantly higher than those in the other five groups(P<0.05).At 5 days of preservation,the mitochondrial mem-brane potential(MMP)of Hu sheep spermatozoa in the 0.02 g/mL fructose-treated group was significantly higher than that in all other groups(P<0.05).Therefore,under the condition of low-temperature preservation,the 0.02 g/mL fruc-tose treatment group had the best effect,effectively prolonging the survival time of spermatozoa and improving the quality of spermatozoa in Hu sheep.关键词
