Abstract

This study investigated the pharmacokinetics and bioavailability of chlortetracycline premix in lamb,provi-ding a scientific basis for clinical research and veterinary prescription formulation.Through in vitro sensitivity tests of E.coli from lambs and treatment trials for lambs with E.coli diarrhea,an effective dose range of chlortetracycline pre-mix was selected.The concentration of chlortetracycline in lamb plasma was measured using high-performance liquid chromatography tandem mass spectrometry,and pharmacokinetics were calculated using DAS 3.0 software.The results showed that for lambs administered a single oral dose of chlortetracycline premix at 12.50,25.00,and 50.00 mg/kg BW every day,the average peak times(Tmax)of its blood chlortetracycline concentration were 10.33,10.67 and 10.67 h,re-spectively;the average maximum blood concentrations(Cmax)were 173.62,420.99 and 642.45 μg/L,respectively;the elimination rate constants were 0.06/h,0.06/h and 0.07/h,with the mean elimination half-life of 12.01,11.77 and 10.50 h,respectively;the average area under the drug-time curve(AUC0-t)were 4 078.70,8 989.64 and 12 412.86 h·μg/L,re-spectively.For the group intravenously injected with chlortetracycline hydrochloride(dosage,11.0 mg/kg BW per day),the elimination rate constant and the elimination half-life were 0.08/h and 9.62 h,respectively,with an AUC0-t of 26 844.91 h·μg/L.The calculated absolute bioavailability of lambs receiving oral doses of chlortetracycline premix at 12.50,25.00,and 50.00 mg/kg BW every day were 13.37%,14.73%,and 10.17%,respectively.In summary,oral ad-ministration of chlortetracycline premix in lambs results in slow absorption,prolonged residence time,slow metabo-lism,and low bioavailability,which is consistent with the requirements for clinical treatment of enteropathogenic E.coli diarrhea in lambs.
