Cloning and Tissue Expression Analysis of CXCL17 Gene in Pamier Yak

Abstract
The CXC family chemokine ligand 17(CXCL17)is a novel chemokine that is mainly expressed in mucosal tissues and has functional roles in maintaining the homeostasis of the mucosal barrier,including regulation of leukocyte recruitment,angiogenesis,and microbial control.In this study,the lung tissue of Pamier yak was used as cDNA tem-plate material,and the sequence of the coding region(CDS region)of the CXCL17 gene was cloned by PCR,and then bioinformatics analysis was performed,and the expression pattern of the gene was analyzed in different tissues,inclu-ding testis,liver,lung,spleen,pancreas,crumpled stomach,cecum,rumen,duodenum and dorsal lumbar longest muscle,by using the qRT-PCR technique.The results showed that the CDS region of the Pamier yak CXCL17 gene was 357 bp in length,encoding 118 amino acids.The protein sequence of Pamier yak CXCL17 was 100.0%homologous to that of the wild yak,followed by 98.3%similarity to that of the cattle.Pamier yak was the closest relative to the wild yak,fol-lowed by cattle and zebu.CXCL17 protein existed as a signal peptide and belonged to hydrophilic proteins.The high-level structure of CXCL17 protein mainly consisted of α-helix(43.22%),irregular coiling(46.61%),and β-folding(10.17%).CXCL17 protein interacted significantly with proteins such as CCL28,CCL25,CXCL10,CXCL14 and CCL26.The CXCL17 gene could be found to have the highest expression in the wrinkled stomach of Pamier yak,fol-lowed by lung,and both were highly significantly higher than that in the other tissues(P<0.01).The lowest expression was in testis.The full CDS region of the CXCL17 gene was successfully cloned in this experiment,and it was disco-vered to be highly expressed in the Pamier yaks'wrinkled stomach and lungs.关键词
