摘要

Abstract

The aim of this study was to investigate the prevalence status and the genetic variation of Haemonchus con-tortus in ovine in Lintao,Gansu province.416 sheep fecal samples were collected from 18 townships in Lintao county using the saturated salt water floating method.ITS-2 gene of Hemoncus contortus was amplified and sequenced by PCR.The base content of ITS-2 sequence was analyzed by DNAStar7.1 software.Moreover,the ITS-2 phylogenetic tree was constructed to analyze the genetic and variation situation of Haemonchus contortus for sheep in Lintao county by using MEGA-X software based on the ITS-2 sequences of 21 isolates from different countries and regions disclosed in GenBank database.The results showed that 126 of the 416 samples were positive for Hemoncus contortus(30.29%);the infection rates in summer and autumn seasons and in winter and spring seasons were 36.05%and 25.24%,respec-tively,with extremely significant differences(P<0.01);the infection rates of young sheep(0-8 months old)and adult sheep(over 9 months old)were 31.95%and 29.63%,respectively,with significant differences(P<0.05).The sequence analysis results showed that the ITS-2 sequences of 126 strains of Hemoncus contortus obtained in this study were all 232 bp in length,and the consistency was 100%,which was named Dingxi strain.The ITS-2 sequence of Dingxi strain showed 95.0%-100%sequence identity with that of Hemoncus contortus isolates from different countries and regions.Phylogenetic tree results showed that the ITS-2 sequence of Dingxi strain was clustered in one branch with those from India(GenBank No.OM334845.1),Chinese Guangdong(GenBank No.KM586652.1)and Ireland(GenBank No.JF680983.1),and they had a close genetic relationship.
