Abstract

In this study,the gene MELO3C002050 of melon was edited by CRISPR-Cas9 technology,aiming to explore the ploidy changes of transgenic melon and its effects on phenotype traits.The phenotype and gene editing of 51 positive seedlings showed that the fruits of 45 plants(type A)were significantly elongated,and the compound gene editing was ex-pected,and the gene editing was successful.6 plants(type B)showed abnormal phenotypes such as enlarged flower or-gans,shortened internodes,shortened fruits,and seed width,but no gene editing occurred.Flow cytometry showed that the DNA content of type B plants was doubled(tetraploid rate of 11.76%).The results of the study showed that during the genetic transformation of melon,it may lead to the spontaneous doubling of ploidy,which may cause phenotypic abnor-malities.Therefore,in the phenotypic evaluation of melon gene editing,ploidy detection is given priority to exclude poly-ploid interference,and it is particularly important to accurately evaluate the phenotype of gene-edited plants.
