Identification of endophytic Rhodotorula mucilaginosa in Cucumis sativus L. and its growth-promoting function

Abstract
To identify cucumber(Cucumis sativus L.)seeds contaminating strains that are frequently present in tissue cul-ture and study their function.In this study,a contaminating strain was isolated and purified,then was classified and identi-fied by a combination of morphology and molecular biology method.A method of reducing the contamination rate of cu-cumber seed tissue culture was explored by heat treatment,meanwhile seed germination test was used to verify its growth-promoting function.The tissue culture contaminant fungi was identified as Rhodotorula mucilaginosa JHJM in cu-cumber seeds.The tissue culture contamination rate of cucumber seeds after strict surface disinfection was 62%,but the rate was reduced to 1%after 8 h heat treatment at 65℃,which was conducive to seed germination,indicating that the fun-gi was endophytes of cucumber seeds.The fermentation broth of the endophyte JHJM significantly promoted the germina-tion of cucumber and peanut,which indicated that the endophyte R.mucilaginosa JHJM could promote seed germination and improve the emergence rate in cucumber cultivation,moreover JHJM could be used as a probiotic preparation into seed coating.As a probiotic to promote seed germination in cucumber cultivation,JHJM causes contamination during tis-sue culture,while the contamination rate could be significantly reduced by heat treatment.This study provides technical support for cucumber tissue culture to reduce pollution and provides strain resources for the preparation of seed coating agent.关键词
