Abstract

The identification of seed purity is imperative to ensure the quality and yield of crops.Conventional field identi-fication methods are inherently limited,and in recent years,molecular marker technologies have been employed in seed purity identification to enhance the accuracy and efficiency of the process.The present study employed three methods-KASP marker,InDel marker and S haplotype type marker based on self-incompatibility to identify the purity of seeds from F1 Chinese cabbage hybrid Yubai CR3.The results demonstrated that among the 188 samples of Yubai CR3,the aforementioned three markers consistently identified two pseudo-hybrids:A4(self-cross of the parent Y636-9)and G2(self-cross of the parent 14CR345M-3).The remaining 186 samples were identified as true Yubai CR3 hybrids,possess-ing alleles of both parents and their purity was 98.94%,thus verifying the consistency and accuracy of the three methods.The study demonstrated that the combined application of multiple molecular marker technologies significantly enhanced the accuracy of seed purity identification,provided substantial technical support for the promotion of Yubai CR3 Chinese cabbage,and also furnished a robust reference for the purity testing of other agricultural seeds.
