Effects and Mechanism of the Compound Puyu Ointment on the Rat Models of Eczema in the Environment at High Altitude

Abstract

Objective In order to analyze the effects and influence on the expression of helper T cell(Th)subpopulation of the compound Puyu ointment on the rat models of chronic eczema in special environment of Xinjiang Plateau.Methods A total of 105 healthy Sprague-Dawley(SD)male rats were divided into seven groups randomly,with 15 rats in each group:blank group,model group,desonide ointment group,low,medium and high concentration of compound Puyu ointment group and vaseline group.In the special environment artificial experimental cabin of northwest,2,4-dinitrochlorobenzene(DNCB)was used to induce the establishment of chronic eczema rat models under high altitude special environment.As the model was successfully established,each group was given corresponding drug.The pathomorphological changes and the eczema area and severity index(EASI)of the rat were observed,the levels of Th1/Th2/Th17/regulatory T cell(Treg)were measured by flow cytometry.Results Except for the model group,vaseline group and control group,the EASI of the rat models in other groups decreased after treatment,the differences were statistically significant(P<0.05).There was a significant statistical difference of the EASI between the high concentration of compound Puyu ointment group and the dexamethasone group in rats after treatment compared with those before treatment(P<0.01),but there was no statistical significance in the difference of efficacy between the two groups(P>0.05).Hyperkeratosis,epidermal elongation,intercellular edema and acanthosis were observed on the model group,which indicated that the model was successful.After drug treatment,the thickening of epidermis,hyperkeratosis and edema of dermis in rat skin tissue were reduced to different degrees.Compared with the blank group,the ratios of Th1 and Treg in the peripheral blood of model rats reduced,the differences were statistically significant(P<0.05).The ratios of Th2 and Th17 increased,the differences were statistically significant(P<0.05).Compared with the model group,the ratios of Th1 and Treg in the peripheral blood of rats increased after seven days treatment with high-concentration compound Puyu ointment,the differences were statistically significant(P<0.05),while the ratios of Th2 and Th17 decreased,the differences were statistically significant(P<0.05).Conclusion Compound Puyu ointment is effective in treating the skin lesions of the rat models of eczema in special environment of Xinjiang Plateau,and its mechanism may be related to inhibiting the inflammatory reaction of skin by regulating the expression of Th subpopulations of the rat models of eczema.
