扶正解毒方对肺炎链球菌肺炎大鼠p38MAPK/NF-κB信号通路的影响
Effects of Fuzheng Jiedu Decoction on p38MAPK/NF-κB Signaling Pathway in Rats with Streptococcus Pneumoniae Pneumonia

Abstract
Objective:To investigate the effects of Fuzheng Jiedu Decoction on inflammatory responses and the p38MAPK/NF-κB signaling pathway in rats with streptococcus pneumoniae pneumonia.Methods:A pneumonia model was established by intratracheal injection of a suspension of streptococcus pneumoniae into the cervical tra-chea of rats.The rats were randomly divided into 6 groups:a blank control group,a model group,low-dose,medi-um-dose,and high-dose traditional Chinese medicine(TCM)groups,and a western medicine group.The respec-tive treatments were administered via gavage for 6 days.Hematoxylin and eosin(HE)staining was used to observe histopathological changes.Blood routine tests were conducted to measure white blood cell counts and differential counts.Enzyme-linked immunosorbent assay(ELISA)was employed to detect levels of IL-6,IL-10,HMGB1,TNF-α inflammatory factors in serum and lung tissues.Western blotting was used to assess protein expression lev-els of p38MAPK,NF-κB p65,and IκBα in lung tissues.Results:Compared with the model group,both Fuzheng Jiedu Decoction and cefuroxime axetil tablets increased white blood cell and lymphocyte counts while reducing neu-trophil counts,as well as levels of IL-6,IL-10,HMGB1,and TNF-α(P<0.05 or P<0.01).They also downregulat-ed protein expression levels of p38MAPK and NF-κB p65 and upregulated IKBα protein expression levels(P<0.05 or P<0.01).Conclusion:Fuzheng Jiedu Decoction can reduce the release of related inflammatory factors in rats with streptococcus pneumoniae pneumonia by inhibiting the activation of the p38MAPK/NF-κB signaling pathway.关键词
