基于Beclin-1泛素化介导巨噬细胞极化探讨益气温阳活血法治疗脓毒症心功能障碍的机制研究
Mechanism of Yiqi Wenyang Huoxue Method in Treating Sepsis-induced Myocardial Dysfunction Based on Beclin-1 Ubiquitination-Mediated Macrophage Polarization

Abstract
Objective:To investigate the mechanism of M2 macrophage polarization in sepsis-induced myocardi-al dysfunction(SIMD)and the therapeutic effect of Yiqi Wenyang Huoxue method.Methods:40 C57BL mice were randomly divided into five groups:a blank control group,an LPS group,low-dose and high-dose Xinyang Tablet groups,with 10 mice in each group.A mouse SIMD model was established by intraperitoneal injection of li-popolysaccharide(10 mg/kg).The survival status of the mice was recorded.After 7 days of treatment,cardiac func-tion indicators,histopathological changes in heart tissue,levels of inflammatory and anti-inflammatory factors,pro-portions of macrophage subtypes,and expression levels of autophagy-related proteins were measured.Results:Compared with the blank control group,LPS-treated mice showed significantly reduced cardiac function(P<0.01),shortened survival time(P<0.001),increased levels of inflammatory factors in myocardial tissue(P<0.05),de-creased levels of anti-inflammatory factors(P<0.01),increased proportion of M1 macrophages,and decreased pro-portion of M2 macrophages.Beclin-1 protein expression and LC3B-Ⅱ/Ⅰ ratio were significantly downregulated(P<0.001).Compared with the LPS group,the Xinyang Tablet groups showed significant improvement in cardiac function indicators,with the high-dose group showing increased survival time,life extension rate,and anti-inflam-matory factor content(P<0.05).In all Xinyang Tablet groups,the proportion of M2 macrophages increased signifi-cantly,while that of M1 macrophages decreased,and Beclin-1 protein expression and LC3B-Ⅱ/Ⅰ ratio were up-regulated(P<0.01).Conclusion:Xinyang Tablet improves cardiac function in SIMD mice by enhancing macro-phage autophagy,promoting polarization from M1 to M2 macrophages,and reducing myocardial inflammation.关键词
