天麻钩藤饮对PTZ诱导癫痫模型小鼠海马神经元的保护及IGF-1R信号通路的调节作用
Protective Effect of Tianma Gouteng Decoction on Hippocampal Neurons in PTZ-induced Epilepsy Model Mice and Its Regulation of the IGF-1R Signaling Pathway

Abstract
Objective:To investigate the protective effect of Tianma Gouteng Decoction on hippocampal neurons in epilepsy model mice through targeting the insulin-like growth factor-1 receptor(IGF-1R)signaling pathway.Methods:40 SPF-grade male mice were randomly divided into four groups:a control group,a model group,a so-dium valproate(VPA)group,and a Tianma Gouteng Decoction group,with 10 mice in each group.An epilepsy model was established by intraperitoneal injection of pentylenetetrazole(PTZ).Each group received corresponding treatments via gavage for 3 weeks.After treatment,hippocampal tissue morphology was observed,seizure condi-tions and cognitive function were recorded,and the expression levels of IGF-1R mRNA,IGF-1R,p-IGF-1R,and Bax proteins,as well as neuronal apoptosis,were detected in brain tissues.Results:In the control group,hippo-campal neurons exhibited normal morphology,closely arranged,with intact and full nuclei,appearing oval-shaped.Compared with the control group,the model group and VPA group showed reduced numbers of hippocampal neu-rons,disorganized and irregular arrangements,nuclear fragmentation,and vacuolated cell structures.Significant dif-ferences were observed among the control,model,VPA,and Tianma Gouteng Decoction groups in terms of EEG to-tal power,average amplitude,and cognitive function(all P<0.05).The expression levels of IGF-1R mRNA in-creased,accompanied by increased apoptosis and elevated Bax,IGF-1R,and p-IGF-1R protein levels(P<0.05).Compared with the VPA group,the Tianma Gouteng Decoction group showed increased numbers of hippocampal neurons,more regular arrangements,alleviated nuclear fragmentation,normalized cell structures,decreased IGF-1R mRNA levels,reduced apoptosis,and lower Bax,IGF-1R,and p-IGF-1R protein levels(P<0.05).Conclu-sion:The protective effect of Tianma Gouteng Decoction on epileptic mice may be achieved by targeting the IGF-1R signaling pathway to inhibit neuronal apoptosis in the hippocampal tissue of epileptic mice.关键词
