苦参碱对急性髓系白血病HL60细胞中Bcl-2/Bax蛋白表达的影响
Effects of Matrine on Bcl-2/Bax Protein Expression in Acute Myeloid Leukemia HL60 Cells

Abstract
Objective:To investigate the effects of matrine on Bcl-2 and Bax protein expression in acute my-eloid leukemia HL60 cells.Methods:HL60 cells were cultured in vitro until the logarithmic growth phase and treated with different concentrations of matrine(0.5,1.0,1.5,2.0 mg/mL)for various durations(0,4,8,24,48 hours).A control group without matrine was also established.Cell viability was assessed using the CCK8 assay,and cell apoptosis was detected by flow cytometry.The expression levels of Bcl-2 and Bax genes were measured using quantitative real-time PCR,and the protein expression levels of Bcl-2 and Bax were determined by Western blotting.Results:The inhibitory effect of matrine on HL60 cell growth was most significant at a concentration of 1.5 mg/mL,with the peak effect observed at 48 hours.Matrine promoted cell apoptosis.Quantitative real-time PCR results showed that in the 1.5 mg/mL matrine treatment group,Bcl-2 gene expression was downregulated,while Bax gene expression was upregulated.Western blotting analysis confirmed these findings,showing that Bcl-2 protein expression was downregulated in the 1.5 mg/mL matrine treatment group,whereas Bax protein expression exhibited an opposite trend.Conclusion:Matrine effectively inhibits HL60 cell viability and accelerates HL60 cell apoptosis,and its mechanism of action may be related to the inhibition of the expression of Bcl-2 and the in-crease of the level of Bax.关键词
