运脾开胃方调控Ghrelin/GHSR/MLCK改善脓毒症大鼠肠黏膜屏障功能的研究
Mechanism of Yunpi Kaiwei Decoction in Regulating Ghrelin/GHSR/MLCK to Improve Intestinal Mucosal Barrier Function in Sepsis Rats

Abstract
Objective:To evaluate the intervention effect of Yunpi Kaiwei Decoction on intestinal mucosal barri-er injury in a sepsis animal model and to explore its mechanism.Methods:60 SD rats were divided into four groups:a sham operation group,a model group,a Yunpi Kaiwei Decoction group,and a Yunpi Kaiwei Decoction+Ghrelin receptor(GHSR)antagonist group.Serum levels of ghrelin,tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6)were measured using enzyme-linked immunosorbent assay(ELISA).Histo-pathological changes in gastrointestinal tissues were observed using hematoxylin-eosin(HE)staining,and intesti-nal epithelial structure was examined by transmission electron microscopy.Intestinal permeability was assessed us-ing the Evans blue method.Protein expression levels of Ghrelin,GHSR,myosin light chain kinase 1(MLCK1),phosphorylated MLCK1(p-MLCK1),myosin light chain(MLC),and phosphorylated MLC(p-MLC)in colon tissue were detected by Western blotting.Results:Compared with the model group,the Yunpi Kaiwei Decoction group showed increased serum ghrelin levels(P<0.01)and decreased pro-inflammatory cytokines TNF-α,IL-1β,and IL-6(P<0.01).After GHSR antagonism,pro-inflammatory cytokine levels increased(P<0.05 or P<0.01).Com-pared with the model group,the Yunpi Kaiwei Decoction group exhibited reduced gastrointestinal pathological changes and decreased intestinal permeability(P<0.01).It also upregulated the expression of Ghrelin and GHSR in colon tissue(P<0.01).However,after GHSR antagonism,the pathological effects were weakened,and intestinal permeability increased(P<0.05).The p-MLCK/MLCK and p-MLC/MLC ratios decreased after Yunpi Kaiwei De-coction intervention,while these ratios increased after GHSR antagonism.Conclusion:Yunpi Kaiwei Decoction can improve intestinal mucosal barrier function in sepsis,and its mechanism may be mediated by the Ghrelin/GH-SR/MLCK signaling pathway.关键词
