Abstract

To establish the somatic embryo regeneration system of Camellia sinensis var.assamica,the cotyledons were used as explants to explore the conditions of explant disinfection,somatic embryo induction and somatic em-bryo seedling formation of 7 resources of Camellia sinensis var.assamica from Yunnan.The results show that the contamination rate was zero under the condition of soaking in 75%ethanol for 1 min and then disinfecting with 0.1%HgCl2 for 16 min.The induction rates of somatic embryos ranged from 0 to 50.84%among 7 resources.The induction rate of 'Linxiang 28' was the highest(50.84%),followed by 'Xiangguiyinhao'(40.09%).Compared with the control group,the addition of 6-BA and 2,4-D significantly increased the induction rate of somatic embryos.The somatic embryo seedling rates of 'Linxiang 28' and 'Xiangguiyinhao' were 21.21%and 1.85%,respectively.This study established a complete somatic embryogenesis regeneration system of Camellia sinensis var.assamica by using 'Xiangguiyinhao' and 'Linxiang 28',which laid a foundation for the preservation of germplasm resources and the construction of genetic transformation system.
