Abstract

The unique geographical environment and climatic conditions of Hangjin Banner have endowed the local Glycyrrhiza uralensis with excellent characteristics such as red skin color,sufficient powder,high acid content and smooth cross-section,making it a typical representative of Ural Glycyrrhiza uralensis in China.The research found that over 50%of the Glycyrrhiza uralensis seeds currently in circulation in the local area were of mixed varieties,and the genetic background was unclear,which brought difficulties to the identifica-tion and protection of genuine Glycyrrhiza uralensis resources.Taking the wild Glycyrrhiza uralensis seeds collected from different geographical populations within Hangjin Banner as the research objects,SNP loci were developed using whole-genome resequencing technology and the Illumina sequencing platform.The obtained sequencing results showed that Q20 was 97.14%to 98.81%,and Q30 was 92.60%to 93.80%.After comparing and screening the sequencing results with the reference genome,a total of 25 772 206 SNP marker sites were obtained.The average value of the secondary allele frequency(MAF)of these SNP variant sites was 0.16,and the average value of the SNP marker polymorphism information content was 0.235.The original SNP loci were screened through multiple indicators,and finally 368 SNP markers capable of identif-ying Glycyrrhiza uralensis in Hangjin Banner from 28 samples were obtained.The corresponding DNA fingerprint maps were constructed and related genetic evolution analyses were conducted.Combined with the error rate results of cross-validation,it was believed that these 28 samples had 2 original ancestors.Three principal components were finally determined from the 28 principal components,namely PC1,PC2 and PC3.
