Abstract

In order to reveal the genetic background of different flower colors of Paeonia lactiflora materials,clarify the genetic relationships among materials,genetic diversity analysis and fingerprint map construction of different flower colors of 56 Paeonia lactiflora germplasm resources were ana-lyzed by using 20 SCoT primers screened in the laboratory.The results showed that a total of 106 bands were amplified by SCoT primers,including 87 polymorphic bands,and the proportion of poly-morphisms was 82.08%;4.35 bands were amplified average per primer,the polymorphic information content ranged from 0.579 0 to 0.891 0,with an average of 0.759 1.The average values of allele num-ber,effective allele number,Nei's gene diversity index and Shannon's information index of different color populations of Paeonia lactiflora were 1.544 8,1.378 0,0.220 0,and 0.244 7,respectively,with the highest values in the light purple population.The genetic similarity coefficients among the five flower colors of Paeonia lactiflora populations were 0.892 6-0.974 5,with the highest genetic simi-larity between the light purple and dark purple populations,indicating a closer genetic relationship.The within-population genetic diversity,the gene differentiation coefficient and the gene flow of dif-ferent flower colors was 0.269 0,0.149 0 and 2.855 1;Variance analysis showed that 96.68%of the genetic variation occurred within different flower color populations.Cluster analysis results showed that 56 Paeonia lactiflora materials were divided into two major groups at a genetic similarity coeffi-cient of 0.68;Fingerprint map of 56 Paeonia lactiflora germplasms was constructed based on 42 genetic loci of 10 SCoT markers.SCoT molecular markers could effectively distinguish Paeonia lacti-flora germplasms with different flower colors,the genetic differentiation among Paeonia lactiflora populations with different flower colors was small.
