Morphological Comparison of Intraspecific Hybrid F1 Generation of Oryzias curvinotus

Abstract
[Objective]To systematically compares the morphological characteristics of hybrid offspring F1 between two geographical populations of Oryzias curvinotus,and analyze the effects of dmy gene expression and geographical population differentiation on the morphological variation and differentiation patterns of hybrid progeny to provide foundation for the taxonomic identification of O.curvinotus.[Methods]Morphological analysis was performed on 300 three-month-old F1 adult fish from six mating groups of O.curvinotus(including three self-crossed groups:ZJ♀×ZJ♂(dmy),ZJ♀×ZJ♂(Lack dmy),and SY♀×SY♂;and three hybrid groups:SY♀×ZJ♂(dmy),SY♀×ZJ♂(Lack dmy),and ZJ♀×SY♂)using traditional morphometric methods.Seventeen measurable traits and five countable traits were systematically measured.Multivariate statistical analyses were applied to compare morphological differences among the groups.For measurable traits,covariance analysis indicated stable heritability of head length(P>0.05).A divergence coefficient(<1.28)confirmed that intraspecific hybridization in O.curvinotus did not reach subspecies-level differentiation.One-way ANOVA further demonstrated no significant differences in head length,caudal fin length,or anal fin length among groups(P>0.05).In countable traits,significant differences were observed in the number of rays in caudal fins,anal fins,and pectoral fins(P<0.05).Hybrid F1 individuals displayed intermediate ray counts between parental phenotypes,except for the ZJ♀×SY♂ group,which exhibited transgressive inheritance(hybrid index<0).Cluster analysis showed morphological divergence between the self-crossed groups ZJ♀×ZJ♂(dmy)and SY♀×SY♂.In contrast,hybrid F1 groups SY♀×ZJ♂(dmy)and ZJ ♀×SY ♂ exhibited phenotypic similarity.These results were corroborated by non-metric multidimensional scaling(NMDS)analysis.Three discriminant function models and principal component analysis(PCA)were established to classify the groups.The overall discrimination accuracies were 67.3%,80.0%,and 62.0%,with cumulative variance contributions of 81.75%,83.02%,and 82.83%,respectively.These results suggest extensive phenotypic overlap(>80%)among groups.[Conclusion]The F1 generation of two geographical populations of O.curvinotus exhibited significant morphological differentiation.All six cross combinations in intraspecific hybridization showed distinct morphological differences in F1 offspring,with paternal effects demonstrating notable influence on phenotypic inheritance.Traditional morphological indicators present challenges in identifying hybrid F1 generations.关键词
