冈村蕨藻多糖的提取优化及其抗氧化和降脂活性
Optimization of Polysaccharide Extraction from Caulerpa okamurae and Evaluation of Their Antioxidant and Hypolipidemic Activities

Abstract
[Objective]To compare the effects of different extraction methods on the bioactivity of Caulerpa okamurae polysaccharide(COP),and screen high-efficiency COP preparation technology,and reveal the synergistic mechanisms underlying its antioxidant and lipid-lowering functions.[Methods]COP was extracted from C.okamurae by four different methods:hot water extraction,ultrasound-assisted extraction,acid extraction,and alkaline extraction.The optimal extraction process was screened based on activity indicators including DPPH and ABTS radical scavenging rates as well as ferric ion reducing power,followed by response surface methodology optimization of critical parameters.Lipid-lowering activity was evaluated through in vitro cholesterol adsorption and bile salt binding assays.FFA-induced non-alcoholic fatty liver disease model in HepG2 cells was established to detect changes in intracellular antioxidant indicators(CAT,SOD and GSH)and lipid metabolism markers(TC,TG and LDL-C).[Result and Conclusion]COP extracted by the hot water showed the better antioxidant effect.The optimized extraction parameters were as follows:the ratio of solid to liquid was 1∶39,the extraction temperature was controlled at 88℃,and the extraction time was 3 h.The lipid-lowering activity of COP was demonstrated by the content of cholesterol and cholate.The activities of catalase,glutathione and superoxide dismutase in the cells were significantly increased,and the contents of total cholesterol,triglycerides and low-density lipoprotein cholesterol were significantly decreased,which proved the antioxidant and lipid-lowering effects of COP in the cell model.These findings provided a theoretical foundation for developing marine green algae-derived functional food additives.关键词
