Exploring the Causal Association Between Gastroesophageal Reflux Disease(GERD)and Meniere's Disease Using Mendelian Randomization

Abstract

Objective Previous studies have shown that Ménière's disease is often accompanied by gastroesophageal reflux disease(GERD).This study aims to assess the potential causal relationship using a two-sample Mendelian randomization(MR).Methods In March 2024,the IEU OpenGWAS platform was searched for genome-wide association studies(GWAS).Single nucleotide polymorphisms(SNPs)strongly associated with GERD were used as instrumental variables(IVs).Five regression models,namely inverse variance weighted(IVW),weighted median(WM),MR-Egger regression,weighted mode,and simple model,were employed to conduct MR analysis on the causal association between GERD and Meniere's disease.Sensitivity analyses were performed using Cochran's Q test,MR Egger intercept test,leave-one-out analysis,and funnel plot.Results IVs indicated increased the risk for Meniere's disease with GERD(OR=1.702,95%CI=1.106-2.618,P=0.01).WM showed similar results(OR=2.205,95%CI=1.214-4.006,P=0.009).Cochran's Q test and funnel plot suggested no heterogeneity among the IVs.MR-Egger intercept test indicated no horizontal pleiotropy in the IVs.Leave-one-out analysis showed that no individual SNP significantly influenced the overall results.Conclusion GERD significantly increases the risk for Meniere's disease.This study provides theoretical evidence for identifying risk factors associated with Meniere's disease.
