摘要

Abstract

[Objective]To establish a high performance liquid chromatography method for L-ascorbic acid 2-glucoside(L-AA-2G)in food.[Method]By comparing and analyzing the mobile phase,chromatographic column,and column temperature in the experimental method,the final mobile phase was determined to be:1.4 g potassium dihydrogen phosphate,26 mL tetrabutylammonium hydroxide(10%)solution,dilu-ted with water to 800 mL,and 89 mL acetonitrile added,mixed well.L-ascorbic acid 2-glucoside was extracted from samples by the mobile phase,was separated by C18 column(4.6 mm×250 mm,5 μm)with temperature of 25℃and ultraviolet detector at 260 nm.[Result]The lin-ear relationship of L-ascorbic acid 2-glucoside was good in the range of 0.5-50.0 μg/mL,with a correlation coefficient(R2)greater than 0.999,the quantification limit was 10.00 mg/kg,the average recovery rate was 95.1%-109.2%,the relative standard deviation(RSD)was 0.065%-2.107%,which met the requirements of experimental detection.[Conclusion]This method has high accuracy and precision,and is used for the determination of L-ascorbic acid 2-glucoside content in solid and liquid food,which could provide a basis for the determination of L-ascorbic acid 2-glucoside content in various food.
