华蟾素对肝癌细胞上皮-间质转化进程的影响
Effect of cinobufagin on the EMT process of hepatocellular carcinoma cells based on molecular docking

Abstract
Objective To investigate the molecular mechanism of cinobufagin(Cinobufagin,Cino)inhibiting the prolifera-tion and migration of hepatocellular carcinoma cells.Methods Molecular docking was used to explore the docking of Cino with Notch1,the core target of Notch pathway.CCK-8 assay was used to detect the effect of Cino on the proliferation of hepatocellular carcinoma MHCC97H cells,and Western blot was used to detect the Notch1 protein expression level.Subsequently,the hepato-cellular carcinoma MHCC97H cells were divided into:the control group,the Cino group,and the Jagged-1(Notch1 agonist)+Cino group.The cell proliferation,migration,and invasion abilities were detected by clone formation,cell scratch,and Tran-swell.The Notch signaling pathway and epithelial-mesenchymal transition(EMT)related protein and mRNA expression levels were detected by Western blot and RT-qPCR.Results Cino docked well with Notch1,the core target of Notch signaling path-way.Cino dose-dependently reduced MHCC97H activity and Notch1 expression in hepatocellular carcinoma cells.Compared with those in the Control group,the mRNA and protein expression levels of Notch1 and Hes1 were significantly reduced in the Cino(2μM)intervention group,the protein expression levels of Vimentin were significantly reduced,and the protein expression levels of E-cadherin were significantly increased,with reduced migration and invasion ability,which was reversed after the intervention of Notch1 agonist Jagged-1.Conclusion Cino can inhibit the proliferation,invasion and migration of hepatocellular carcinoma MHCC97H cells,and its mechanism of action is related to the reduction of the activation level of Notch signaling pathway,which provides a new idea for the study of anti-tumor effects of traditional Chinese medicine.关键词
