细胞爬片式共培养模型的建立和优点
Establishment and advantages of cell crawing sheets co-culture model

Abstract
Objective To investigate the effects of human lung adenocarcinoma A549 cells in different co-culture modes on the phenotype and function of human umbilical vein endothelial fusion cells EA.hy.926 cells.Methods The co-culture models of A549 cells and EA.hy.926 cells were established by "cell creep" co-culture method and "transwell chamber" co-culture method,respectively,and the integrity of endothelial cell barrier was evaluated by FITC-glucan cell penetration test.The pheno-type and functional changes of EA.hy.926 cells were detected by Western blot analysis of polysaccharide coated core proteins SDC-1,tight junction proteins ZO-1,occludin-related proteins and CCK8 detection of endothelial cell proliferation activity.Re-sults Compared with transwell cell co-culture model,in the FITC-glucan cell penetration test,the infiltration rate of EA.hy.926 cells was significantly increased by cell creep co-culture model(P<0.05),indicating that endothelial cell integrity was damaged and cell permeability was increased;by Western blot,the cell creep co-culture model significantly downregulated the expression of SDC-1,ZO-1 and Occludin of EA.hy.926 cells,and the damage was obvious(P<0.05);by CCK8 experiment,the prolifer-ation activity of EA.hy.926 cells was significantly increased in the creep co-culture model(P<0.05).Conclusion Compared with transwell cell co-culture model,the cell creep co-culture model can better reflect the influence of tumor cells on endothelial cells,providing a new research model for the study of lung cancer metastasis.关键词
