Abstract
Objective To explore the mechanism of noncanonical NF-κB signaling pathway mediated by B-cell activation fac-tor(BAFF)and nuclear factor kappa-B(BAFF-R)in antiphospholipid syndrome.Methods Blood samples were collected from primary antiphospholipid syndrome(APS)patients and healthy adults,and peripheral blood single nucleated cells were separated by human lymphocyte isolation solution.They were divided into the APS group and the healthy control group,and cells were cul-tured and collected.The levels of BAFF and related inflammatory factors were detected by ELISA;the expression levels of BAFF-R were analyzed by flow cytometry;and Western blot method was used to determine the expression levels of tumor necrosis factor receptor 2(TNFR2),TNF receptor associated factors(TRAF3),inhibitor of kappa B kinase(IKK),p100,p52,NF-κB pro-tein,IL-1β and IL-6.Results BAFF and BAFF-R levels in the APS group were higher than those in the healthy control group(P<0.01).The protein expression levels of TRAF2,IKK,p52,and NF-κB in the APS group were higher than those in the healthy control group(P<0.01),and the protein expression levels of TRAF3 and p100 were lower than those in the healthy control group(P<0.01).The expression levels of IL-1β and IL-6 factors in the APS group were higher than those in the healthy control group(P<0.01).Conclusion The noncanonical NF-κB signaling pathway mediated by BAFF and BAFF-R is involved in cell activation and stimulates the production of inflammatory factors.关键词
