Construction of a prognostic model of key genes for sorafenib resistance in hepatocellular carcinoma

Abstract

Objective To investigate the key genes associated with prognostic resistance to sorafenib in primary hepatocellular carcinoma(HCC)by applying bioinformatics technology to explore new therapeutic avenues and address drug resistance.Meth-ods LIHC gene expression data from the GSE109211 dataset and TCGA and ICGC databases were downloaded from the GEO da-tabase to screen for prognostic differential genes for sorafenib resistance in liver cancer.The biological functions of HCC sorafenib resistance differential genes were analyzed by using R software.Results Two hundred and one genes significantly associated with prognostic drug resistance were screened out,which were mainly involved in small molecule biosynthesis,amino acid metabolism and other biological processes.Five optimal model genes,ZNF831,FTCD,KIF4B,CCT3 and SpINK5,were obtained,a risk score model was constructed based on these genes,and the samples were divided into high-risk and low-risk groups according to the median risk score.The results of the study showed that the high-risk group had shorter survival time(P<0.001)and higher sensitivity to chemotherapeutic agents(P<0.001),and both tumour stage and risk score were independent prognostic factors.Based on the prognostic drug resistance gene expression data,LIHC tumour samples were classified into two subtypes,C1 and C2,and the survival rate of the C2 subtype was significantly higher than that of the C1 subtype(P<0.001).Conclusion Risk-Score model constructed based on the prognostic key genes for sorafenib resistance in HCC has the potential to provide important predictions of prognosis,immune microenvironment and chemotherapeutic response in patients with primary HCC.
