Abstract

To investigate the effects of different sources and preparation processes on the antioxidant proper-ties of natural melanin nanoparticles,squid ink sacs and yak wool were used as raw materials,three preparation processes such as deionized water extraction,alkaline ultrasonic rotary evaporation and acidic reflux were em-ployed to prepare water-extracted squid melanin(WM),water-soluble squid melanin(WSSM)and yak mela-nin(YM)respectively.The morphology,structure and scavenging properties against hydroxyl radicals(·OH),superoxide radicals(O2·)and hydrogen peroxide(H2O2)of the three types of melanin were analyzed.And the influence of particle size on the antioxidant properties was investigated.The results showed that different prep-aration methods could affect the aggregation degree of melanin monomers.The more severe the reaction condi-tions,the smaller the aggregation degree among melanin monomers,and more monomers could react with free radicals.The aggregation degree of melanin monomer in WM was the highest and the dispersion was the worst,followed by YM,while the aggregation degree in WSSM was the lowest with the best dispersion.WSSM showed the best antioxidant property and its·OH scavenging ability was 38%higher than WM and 18%higher than YM.Particle size analysis exhibited that the average particle sizes of WM,YM and WSSM were 630 nm,501 nm and 342 nm respectively,which further confirmed that WSSM had better dispersibility and could react with free radicals more effectively.The study results have important reference value for the application of melanin in medi-cal dressing field.
