Role of SIRT3 in Children with Acute Myeloid Leukemia

Abstract
Objective To explore the effect of silent information regulator 3(SIRT3)in chil-dren with acute myeloid leukemia(AML).Methods The expression level of SIRT3 in bone mar-row cells of AML children and peripheral blood leukocytes of healthy blood donor children was de-tected.SIRT3 expression was knocked down in AML cells.The cells' proliferation,apoptosis,oxi-dative stress indexes,and protein expression levels of the Wnt/β-catenin signaling pathway were detected.Results Compared with those in the normal children group,the expression levels of SIRT3 mRNA and protein were increased in the AML children group(P<0.05).After knocking down SIRT3 expression in AML cells,the cells proliferation,the colony formation ability,the levels of glutathione(GSH)and superoxide dismutase(SOD),the total antioxidant capacity(T-AOC),the expression level of β-catenin,and the transcription activity of T cell factor/lymphocyte enhancer factor(TCF/LEF)were decreased,while the apoptosis rate,the levels of reactive oxygen species(ROS),malond-ialdehyde(MDA)and phosphorylated glycogen synthase kinase 3β(p-GSK-3β)were increased(P<0.05),the GSK-3β inhibitor TWS119 could partially reverse the above effects.Conclusion Knoc-king down SIRT3 can inhibit the growth of AML cells and increase oxidative stress level,which may be related to the inhibition of Wnt/β-catenin signaling pathways.关键词
