Traditional Chinese Medicine Preventing and Treating Renal Fibrosis by Regulating Autophagy Mediated by TGF-β Pathway

Abstract
TGF-β signaling pathway is the main pathway that regulates autophagy,which can promote the progression of renal fibrosis by mediating autophagy.Autophagy is a mechanism that tends to homeostasis in the intracellular environment and is widely involved in multiple aspects of renal fibrosis.Traditional Chinese medicine monomers and compounds can directly act on targeted cells and autoph-agy targets with multiple targets and multiple effects,and play a key role in the prevention and treatment of renal fibrosis.However,there are still some limitations in the research on the treatment of renal fibrosis by targeting TGF-β-mediated autophagy in traditional Chi-nese medicine:(1)The development of renal fibrosis is not a static pathological process,and in addition to TGF-β-mediated autoph-agy,there are other related cytokines and signaling pathways that work together to promote the progression of renal fibrosis.(2)There are still many unknown mechanisms of autophagy that need to be explored,and there are two issues worthy of attention:① most studies have not analyzed how the specific active components of related TCM monomers are involved in the regulation of autophagy;(2)The role of microautophagy and chaperone-mediated autophagy in different types of renal cells in renal fibrosis is not yet known.(3)In vitro model studies of TCM intervention are the mainstay,and there is a lack of relevant evidence-based evidence.Therefore,network phar-macology,Mendelian randomization studies and other analyses were used to explore the targets of specific active ingredients of TCM monomers and compounds,and corresponding high-quality and randomized controlled clinical experiments were carried out to ensure the effectiveness of TCM.关键词
