背部推法通过调节色氨酸-犬尿氨酸代谢途径改善大鼠慢性疲劳综合征认知障碍
Tuina Manipulating on Back Improving Cognitive Impairment in Chronic Fatigue Syndrome in Rats by Regulating the Tryptophan-Kynurenine Metabolism Pathway

Abstract
Objective:To investigate the mechanism of Tuina manipulating on back in improving cognitive impairment in rats with chronic fatigue syndrome(CFS)by regulating the tryptophan-kynurenine pathway(KP).Methods:8 SD rats were randomly selected as the blank group,and the rest of the rats were divided into model group,push group and 1-methyl-L-tryptophan(1-MT)group,with 8 rats in each group.After 21 days of modeling,the 1-MT group was given 1-MT solution by gavage,and the Tuina group operated the push-up method on the back of the rats once a day for 14 consecutive days.The open field experiment was used to detect the motor function of rats,and the new object recognition experiment was used to detect the cognitive function of rats.ELISA was used to detect in-terleukin(IL)-1β,IL-6,tumor necrosis factor-α(TNF-α),kynurenic acid(KYNA),kynurenine-3-monooxygenase(KMO),3-3-hydroxy-kynurenine(3-HK)and quinolinic acid(QUIN)content.Western blot was used to detect the protein expression level of N-methyl-D-aspartate receptor(NMDAR)in the hippocampus,and the positive percentage of ionized calcium binding a-dapter molecule-1(Iba-1)in the hippocampus was detected by immunofluorescence.Results:Compared with the blank group,the total distance travel,distance in the center area,time in the center area,number of entries in the center area and number of cross-grids in the model group were reduced(P<0.05).Compared with the model group,the total distance,distance in the central area,time in the central area,number of entries in the central area and the number of spans in the 1-MT group were increased(P<0.05).The new object recognition experiment showed that compared with the blank group,the recognition index and recognition probability of the rats in the model group were reduced(P<0.01).Compared with the model group,the recognition index and recognition probability of the rats in the push-method group and the 1-MT group were increased(P<0.01).Compared with the blank group,the levels of IL-1β,IL-6,TNF-α,KMO,3-HK,and QUIN in the hippocampus of the model group rats increased,while the level of KYNA de-creased(P<0.01);Compared with the model group,the levels of IL-1β,IL-6,TNF-α,KMO,3-HK,and QUIN in the hippo-campus of rats in the Tuina group and 1-MT group decreased,while the level of KYNA increased(P<0.05).Compared with the blank group,the protein expression levels of NMDAR1,NMDAR2A,and NMDAR2B in the hippocampus of the model group rats in-creased(P<0.01);Compared with the model group,the expression levels of NMDAR1,NMDAR2A,and NMDAR2B proteins in the hippocampus of rats in Tuina group and 1-MT group were reduced(P<0.01).Compared with the blank group,the positive percent-age of Iba-1 in the hippocampal CA1 region of the model group rats increased(P<0.01);Compared with the model group,the per-centage of Iba-1 positivity in the hippocampal CA1 region of rats in Tuina group and 1-MT group decreased(P<0.01).Conclu-sion:Tuina manipulating on back method may improve cognitive impairment in CFS rats by regulating the KP pathway and inhibiting neurotoxicity.关键词
