Differential Analysis of Active Metabolites in Different Jujube Flower Varieties Based on UHPLC-Q-TOF MS

摘要

Abstract

To explore the diversity and potential differences in biological activities of active metabolites across various jujube varieties,and provide a theoretical foundation for improving jujube cultivars and enhancing their medicinal and edible value,three primary cultivated varieties of jujube flowers from Shanxi Province,namely Banzao(BZ),Dongzao(DZ),and Hupingzao(HP),were utilized as experimental materials.Ultra-high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry and multivariate statistical methods were employed to analyze the metabolomic profiles of jujube flowers.The results revealed that a total of 178 metabolites were identified,among which 108 were classified as active metabolites,accounting for 60.67%of the total identified compounds.These active constituents primarily included polyphenols,organic acids,amino acids,sugars,vitamins,and terpenoids.Significant variations were observed among different jujube flower samples.With screening criteria of variable importance projection(VIP)>1 and P<0.05,46 significantly different active metabolites were identified,comprising 26 up-regulated and 20 down-regulated substances.Cluster heatmap analysis indicated distict expression patterns of active metabolites across jujube flower varieties.Specifically,ten metabolites,such as squalene and vanillic acid,exhibited significant up-regulation in Banzao jujube flowers;whereas nine unique metabolites,including resveratrol and isoquercetin,displayed enhanced expression levels in Dongzao and Hupingzao jujube flowers.Furthermore,KEGG pathway enrichment analysis highlighted arginine and proline metabolism and phenylpropanoid biosynthesis as the principal metabolic pathways shared by all three types of jujube flowers.However,key metabolites such as agmatine,4-agmatinoic acid,L-glutamic acid,and 4-nitrophenol demonstrated variability among groups.The results of this study offer a theoretical foundation for comprehensive investigating active metabolites in jujube flowers,and for developing diverse jujube flower resources.
