Abstract

Aim To explore the effect of knocking down BASP1 on autophagy in human embryonic kidney 293(HEK293)cells based on bioinformatics.Methods The correlation between BASP1 and human diseases was analyzed using bioinformatics.BASP1-interacting protein were screened by ultra-high performance liquid chromatography-mass spectrometry,and the enriched path-ways of related genes were analyzed by using KEGG and GO.After cells were divided into experimental groups,the autophagy related protein levels of LC3,p62,and ulk1 in each group were detected by immunoblotting,and changes in autophagic structure of cells in each group were observed by using transmission electron microscopy.Results Bioinformatics analysis indicates that the expression level of BASP1 is associated with diseases such as Alzheimer's disease and tumors.The proteins interacting with BASP1 are mainly enriched in the pathways of neurodegenerative diseases and the process of cellular autophagy.The cell experiment results showed that knocking down BASP1 increased the proportion of autophagic lysosomes and the level of ulk1 protein(P<0.05);After treatment with the autophagy flow blocker chloroquine,knocking down BASP1 resulted in a decrease in the proportion of autophagosomes,an increase in the proportion of autophagosomes,and an increase in the expression level of LC3 protein(P<0.05).Conclusion Knockdown of BASP1 can enhance the autophagic flow of HEK293 cells,revealing its association with cellular autophagy.
