过表达miR-9-5p对槟榔碱诱导BMF活化能力的影响机制

Abstract
Aim To explore the mechanism of the effects of overexpression of miR-9-5p on the activation ability of oral buc-cal fibroblast(BMF)cells.Methods Different concentrations(0,5,10,20 μg/mL)of arecoline were applied to BMF to in-duce activation of myofibroblasts(fBMF),and the optimal stimulation concentration was screened.BMF cell experiment was divided into control group,arecoline+inhibitor control(i-NC)group and arecoline+miR-9-5p inhibitor(i-miR-9-5p)group,respectively.The fBMF cell experiment was divided into negative control(NC)group,miR-9-5p mimic(miR-9-5p)group,i-NC group,i-miR-9-5p group,i-miR-9-5p+RUNX3 lentivirus negative control(sh-NC)group and i-miR-9-5p group,respectively.The expression of miR-9-5p gene was analyzed by real-time PCR.Cell migration test and wound healing test were used to evaluate cell migration ability.The expression levels of MAPK signaling pathway related proteins were analyzed by using Western blotting.Results 10 mg/L arecoline was the best concentration for BMF cells.Compared with the control group,the contraction of collagen gel in the arecoline+i-NC group was decreased significantly,and the trans pore migration and wound healing were significantly increased.Compared with the arecoline+i-NC group,the contraction of collagen gel in the arecoline+i-miR-9-5p group was significantly increased,and the trans pore migration and wound healing were significantly decreased.Compared with the i-NC group,the collagen gel contractility of fBMFs in the i-miR-9-5p group was significantly increased,and the migration and wound healing ability of fBMFs were significantly decreased.miR-9-5p inhibited RUNX3 expression and enhanceed myofibroblast activation by directly targeting RUNX3.The interaction be-tween miR-9-5p and RUNX3 enhanced myofibroblast activation by inducing the MAPK pathway.Conclusion Overexpression of miR-9-5p enhances myofibroblast activation by inhibiting RUNX3,which may be related to MAPK pathway activation.关键词
