摘要

Abstract

Aim To investigate the molecular mechanism of Tongluo Yishen Formula(TLYS)in ameliorating renal fibrosis through transcriptomics and network pharmacology.Methods The unilateral ureteral obstruction(UUO)rat model was established and divided into the model group,TLYS treatment group,and sham-operated group.After 4 weeks of drug administra-tion,kidney tissues were collected for transcriptomic sequencing and enrichment analysis.Potential targets of TLYS active components were retrieved from TCMSP,Swiss Target Prediction,PubChem,and protein-protein interaction(PPI)databases(STRING).Renal fibrosis-related targets were obtained from GeneCards.A PPI network was constructed for common targets be-tween TLYS components and renal fibrosis,followed by enrichment and network topology analyses.Results Sequencing revealed 295 differentially expressed genes(200 upregulated,95 downregulated)in the TLYS group compared to the model group,with DUSP15 and CTXN3 showing the most significant changes.Functional enrichment analysis indicated that TLYS exerted anti-fibrotic effects by regulating extracellular matrix(ECM)remodeling,protein binding activity,and the TGF-β signaling pathway,while also implicating TRPC channel-mediated calcium signaling,PI3K-Akt pathway activation,and inflammatory cytokine network homeostasis.Network pharmacology identified 16 overlapping targets between TLYS components and renal fibrosis,primarily enriched in ECM regu-lation and immune response.PPI network analysis further suggested Salvia miltiorrhiza(Danshen)as a key component of TLYS in modulating the immune microenvironment.Pathway analysis demonstrated that TLYS mechanisms were closely associated with lysoso-mal metabolic pathways and apoptotic signal transduction.Conclusion TLYS effectively mitigates renal fibrosis by targeting ECM deposition,apoptotic pathways,and renal inflammation through a multi-component,multi-target approach,offering potential for pre-vention and early intervention of fibrosis.
