Abstract
Aim To investigate the feasibility of using the optimized Blue-White spot screening method to evaluate the PCR detection system for epidermal growth factor receptor(EGFR)gene deletion mutation.Methods An optimized Blue-White spot screening method was constructed to evaluate the performance of PCR detection systems for assessing exon 19 deletion mutations in EG-FR and cell free DNA sample PCR-Blocker primer detection systems.Using wild-type plasmids,mutant plasmids,genomic DNA from blood,and peripheral blood free DNA as templates,the detection results were determined by colony color.Results In the detection of EGFR gene exon 19 deletion mutation,wild-type colonies were white,and mutant colonies were blue.The ratio of blue to white colonies from different templates reflected the system's specificity and sensitivity.The ratio of blue to white colonies in the PCR-Blocker primer system was significantly higher than that in the PCR detection system(P<0.05).Conclusion The optimized Blue-White spot screening method can effectively evaluate the performance of EGFR deletion mutation PCR detection system.关键词
