PLX3397对LPS所致小鼠抑郁样行为的影响
The effects of PLX3397 on depressive-like behavior in mice induced by LPS

Abstract
Aim To investigate the effects of the colony stimulating factor 1 receptor inhibitor pexidartinib(PLX3397)on lipopolysaccharide(LPS)-induced depressive-like behaviors in mice.Methods Thirty-six mice were randomly divided into the normal group,the model group,and the PLX3397 group.Depressive-like behavior in each group of mice were evaluated by sugar wa-ter preference experiment,open field experiment,suspended tail experiment,and forced swimming experiment.The spleen indices of each group were compared.Western blotting was used to detect the expression levels of the hippocampal microglial cell marker ion-ized calcium-binding adapter molecule 1(IBA1)in each group,and PCR was used to detect the mRNA expression levels of IBA1 and inflammatory factors interleukin(IL)-1[3,IL-6,IL-1α,and tumor necrosis factor(TNF)-α in the hippocampus of each group.Results Compared with the normal group,the model group and PLX3397 group showed a decrease in the percentage of sugar water preference,total exercise distance,and central area time in the open field experiment(P<0.05);The time of hanging tail immobility and forced swimming immobility increased(P<0.05).There was no significant difference in the above indicators between the model group and the PLX3397 group(P>0.05).Compared with the normal group,the model group showed an increase in the expression levels of IBA1 protein and its mRNA,as well as IL-1β,IL-6,IL-1α,and TNF-α mRNA(P<0.05);Compared with the model group,the PLX3397 group showed a decrease in the levels of the above indicators(P<0.05).Conclusion PLX3397 can elimi-nate the depletion of hippocampal microglia and alleviate LPS-induced neuroinflammation,but does not alleviate LPS-induced depres-sive-like behavior in mice.关键词
