积雪草苷脂质体凝胶促进糖尿病足大鼠创面愈合的作用及机制
Effect and mechanism of asiaticoside liposome gel on promoting wound healing in rats with diabetic feet

Abstract
Aim To explore the effect and mechanism of asiaticoside liposome gel on promoting wound healing in rats with diabetic feet ulcer(DFU).Methods SD rats were randomly divided into control group,model group,centella triterpenes cream group and liposome gel group.Except control group,DFU models were constructed in the other groups.The body mass,blood glu-cose and wound healing rate in different groups were compared.The pathological changes of wound tissues were observed by HE stai-ning,expression level of CD31 was detected by immunohistochemistry,levels of superoxide dismutase(SOD)and malondialdehyde(MDA)were detected by the kit,and expressions of nuclear factor erythroid 2-related factor 2(Nrf2),heme oxygenase-1(HO-1)and NQO1 proteins were detected by Western blotting.Results After modeling,body mass was increased in control group but de-creased in model group,centella triterpenes cream group and liposome gel group,and body mass in the three groups was significantly lower than that in control group at 14 d and 21 d.The blood glucose in model group,centella triterpenes cream group and liposome gel group was higher than that in control group at different time points(P<0.05).The wound healing rate was gradually decreased in control group,liposome gel group,centella triterpenes cream group and model group,and was higher at 14 d after administration than that at 1 d after administration in all group(P<0.05).The pathological changes of wound tissues were the most significant in lipo-some gel group.In control group,liposome gel group,centella triterpenes cream group and model group,positive expression rate of CD31,SOD activity,expression levels of Nrf2,HO-1 and NQO1 were gradually decreased,while MDA was gradually increased(P<0.05).Conclusion Asiaticoside liposome gel can promote wound healing in DFU rats by activating Nrf2 pathway.关键词
