Abstract

To address the question,which is the mechanism that alkyl organophosphorus ester(OPEs)-toxicity on phytoplankton growth is remain largely unknown.This study chose tributyl phos-phate(TnBP)was chosen as a representative of alkyl OPEs to investigate its effects on the growth of marine phytoplankton,Phaeodactylum tricornutum,and to reveal in deep its potential toxic mechanism based on the changes of key genes of photosynthesis and energy metabolism,with a view to establishing a theoretical reference for the model of the toxic effect of alkyl OPEs on phytoplankton.The results showed that the population growth of Phaeodactylum tricornutum was significantly inhibited when ex-posed to TnBP at concentrations of 1.5(the maximum level in aquatic environment),3.0 and 4.5 μmol/L,and the intracellular total organic carbon,chlorophyll,and carotenoids contents were also reduced.The chlorophyll fluorescence detection showed that TnBP reduced the maximum photochemical rate Fv/Fm of photosystem Ⅱ and non-photochemical quenching capacity in P.tricornutum,which indicated that photosynthesis was significantly inhibited.Real-time fluorescence quantitative PCR re-sults showed that TnBP down-regulated the expression of two key genes for chlorophyll synthesis,which encoded coproporphyrinogen Ⅲ oxidase and the D subunit of magnesium chelate.Meanwhile,the expression level of two genes(ZEP1 and ZEP3)that encode zeaxanthin epoxidase,a key enzyme for lutein synthesis,also showed a declining trend.It indicated the ability of light-capture and photoprotec-tion of algal cells impaired,which led to a decrease in photosynthetic efficiency and a reduction in organ-ic carbon content.Additionally,it was found that the genes encoding succinate dehydrogenase SDH2 and glyceraldehyde-3-phosphate dehydrogenase GAPC1 were down-regulated,which was suggested that this may lead to an nttenuation of glycolytic pathway and tricarboxylic acid cycle within the algal cells.Therefore,TnBP interfered with intracellular substance synthesis and accumulation by phytoplankton photosynthesis and energy metabolism,which ultimately inhibited cell division and growth.The results of this study can provide a theoretical reference for the establishment of the toxic effect model of alkyl OPEs on phytoplankton.
