Two Strains of Marine Origin Comparative Genomic Analysis of the Genus Mycolicibacterium sp.

Abstract
In this study,two strains of Mycolicibacterium sp.LZB054 and YHM108 isolated from the East China Sea and the Mariana Trench,respectively,and their related strains were studied by compara-tive genomic analyses to reveal their genetic characteristics and metabolic potential.The genome se-quences of LZB054 and YHM108 were obtained by whole genome sequencing,and their taxonomic sta-tus was determined by phylogenetic analysis based on 16S rRNA genes and single-copy core genes.Functional annotation and comparative genomic analysis of these two strains and their reference strains were performed to reveal the genomic characteristics and metabolic potentials of the genus.The genome sizes of strains LZB054 and YHM108 were 6 150 419 and 6 164 777 bp,with GC contents of 67.99%and 67.87%,and 5 761 and 5 848 encoding genes,respectively.Phylogenetic analysis showed that strains LZB054 and YHM108 were homozygous,but the DNA-DNA hybridization values with their clo-sest strains were less than 70%,which indicated that they were potentially new species in the genus Mycolicibacterium.Functional annotation analysis revealed a relatively high percentage of genes with functions of lipid transport and metabolism,transcription,and secondary metabolite biosynthesis.Strain LZB054,isolated from the East China Sea,contained a gene coding for Fe-Mn transporter MntH,while strain YHM108 from the Mariana Trench contained a gene coding for colanic acid and biofilm tran-scriptional regulator,formic acid oxidation and methane oxidation functions.In addition,these two strains were annotated with a variety of carbohydrate hydrolases such as glucosidase and phosphatidyli-nositol mannosyltransferase,and were predicted to have more than 10 clusters of secondary metabolite synthesizing genes,which indicated that these two strains might have a very high potential development value.The genetic characteristics and metabolic potential of two novel strains of Mycolicibacterium sp.LZB054 and YHM108 were revealed through comparative genomic analysis,which provided data sup-port and theoretical basis for the further study of their adaptation mechanism to marine environment and the future development and utilization of the strains.关键词
