Regulatory Roles of ALKBH5 in Tumorigenesis and Tumor Immunity:Mechanisms and Drug Development
RNA 去甲基化酶 alkB 同源物 5(alkB homolog 5,ALKBH5)介导的 N6-甲基腺苷(N6-methyladenosine,m6A)去甲基化通过影响RNA代谢的多个途径调节基因的表达.研究发现,ALKBH5在许多恶性肿瘤的进程中以m6A依赖性方式发挥关键调控作用.因此,探究ALKBH5在肿瘤中发挥的生物学功能和机制,对攻克人类恶性肿瘤和研发相关靶向药物具有重要意义.本综述重点阐述了ALKBH5在肿瘤中发挥各种生物学功能的分子机制和其在肿瘤相关免疫中的作用,并且讨论了靶向ALKBH5的肿瘤治疗策略及临床意义,以期为ALKBH5介导肿瘤发生发展分子机制的深入研究和相关防治药物的研发提供新的思路和见解.
The RNA demethylase alkB homolog 5(ALKBH5)regulates gene expression by mediating N6-me-thyladenosine(m6A)demethylation through multiple pathways of RNA metabolism.Studies have demonstrated that ALKBH5 plays a critical regulatory role in the progression of various malignancies in a m6A-dependent manner.Therefore,investigating the biological functions and mechanisms of ALKBH5 in tumors holds con-siderable importance for combating human malignancies and developing targeted therapies.This review fo-cuses on the molecular mechanisms by which ALKBH5 exerts diverse biological functions in tumors,its roles in tumor-related immunity,and ALKBH5-targeted therapeutic strategies along with their clinical implica-tions,with aiming to provide novel insights and perspectives for in-depth research on ALKBH5-mediated molecular mechanisms in tumorigenesis and development of related therapeutic agents.
