基于Cathepsin-B/NLRP3信号通路探讨娄氏清痹汤治疗急性痛风性关节炎的作用机制
Exploring the mechanism of Loushi Qingbi Decoction(娄氏清痹汤)in treating acute gouty ar-thritis based on the Cathepsin-B/NLRP3 signaling pathway

Abstract
Objective To investigate the effects and mechanism of Loushi Qingbi Decoction(娄氏清痹汤,LSQBD)on acute gouty ar-thritis(AGA)model rats.Methods The AGA model was established using monosodium urate(MSU)suspension.LSQBD was adminis-tered for 5 days before modeling,and samples were collected 48 hours after modeling.Ankle swelling degree and gait scores were per-formed.Hematoxylin-eosin(HE)staining was used to detect synovial tissue morphology changes of ankle.ELISA and biochemical as-says were employed to measure serum inflammatory factors and oxidative stress levels of rats in each group.RT-qPCR,immunohisto-chemistry(IHC),and Western blot(WB)were used to detect mRNA and protein expressions of the Cathepsin-B/NLRP3 signaling pathway in the synovial tissues.Results Compared with the blank control group,the model group exhibited typical acute inflammatory re-sponses,significantly increased ankle swelling and gait scores(P<0.01),with synovial tissue inflammation of massive inflammatory cell infiltration and capillary hyperplasia and elevated histopathological scores(P<0.01).Compared with the model group,LSQBD and colchicine groups showed significant improvements in ankle swelling and gait scores(P<0.05,P<0.01),alleviated inflammatory cell infiltration and hyperplasia,decreased histopathological scores.Serum levels of IL-1β,TNF-α,IL-6,and ROS,MDA were signifi-cantly reduced(P<0.05,P<0.01),while SOD,GSH,and CAT levels were increased(P<0.05,P<0.01).Additionally,mRNA and protein expression levels of the Cathepsin-B/NLRP3 pathway in the synovial tissues was down-regulated(P<0.01).Conclusion LSQBD can alleviate acute inflammatory responses in AGA,and its action mechanism may be associated with the modulation of the Ca-thepsin-B/NLRP3 signaling pathway.关键词
