摘要

Abstract

Objective To investigate the effects of Maimendong(Radix Ophiopogonis)Decoction(麦门冬汤,MMDD)on bleomycin(BLM)-induced pulmonary fibrosis(PF)in rats based on the Hippo signaling pathway.Methods Rats were randomly divided into 6 groups.A PF model was established by intratracheal injection of BLM,and samples were collected at 14 and 28 days after modeling.He-matoxylin-eosin(HE)staining,Masson staining,immunohistochemistry(IHC)and RT-qPCR were used to detect the morphological changes in the lung tissues and the levels of MST1,TGF-β1,Smad3 and Yap.Results HE and Masson staining revealed varying de-grees of pathological changes in the lung tissues across all groups compared to the blank group.After drug intervention,the morphological structure and fibrosis severity in the treatment groups were alleviated to varying degrees compared to the model group.IHC and PCR re-sults showed that,compared to the blank group,the protein and mRNA expression of MST1 were decreased at both time points,while the protein and mRNA expressions of TGF-β1,Smad3,and Yap were increased in varying degrees(P<0.05).Compared to the model group,the treatment groups exhibited increased MST1 expression and decreased protein and mRNA expressions of TGF-β1,Smad3,and Yap(P<0.05).Conclusion MMDD can ameliorate the progression of PF by regulating the Hippo signaling pathway.
